Production of xylanolytic enzymes by anAspergillus niger CCMI 850 isolate was investigated in batch cultures. The effect of the composition of a fermentation medium that did not include chemical inducers, on β-xylanase, β-xylosidase, α-l-arabinofuranosidase, and total cellulase activity was studied. With 4% xylan as the carbon source, about 65 U/mL of β-xylanase was obtained, whereas the total cellulase activity was undetectable, under the specified conditions. This β-xylanase activity represents the highest reported for a wild-type strain ofA. niger. The effect of pH and temperature on the activity of β-xylanase was studied. Partial characterization of the β-xylanase showed that with insoluble birchwood as substrate theK m andV max were 0.3 mM and 19 μmol/min, respectively. Aspects of using the crude β-xylanase preparation for applications in the pulp and paper industry were discussed.  相似文献

Fast determination of neighboring atoms is an essential step in molecular dynamics simulations or Monte Carlo computations, and there exists a variety of algorithms to efficiently compute neighbor lists. However, most of these algorithms are general, and not specifically designed for a given type of application. As a result, although their average performance is satisfactory, they might be inappropriate in some specific application domains. In this article, we study the case of detecting neighbors between large rigid molecules, which has applications in, e.g., rigid body molecular docking, Monte Carlo simulations of molecular self-assembly or diffusion, and rigid body molecular dynamics simulations. More precisely, we compare the traditional grid-based algorithm to a series of hierarchy-based algorithms that use bounding volumes to rapidly eliminate large groups of irrelevant pairs of atoms during the neighbor search. We compare the performance of these algorithms based on several parameters: the size of the molecules, the average distance between them, the cutoff distance, as well as the type of bounding volume used in the culling hierarchy (AABB, OBB, wrapped, or layered spheres). We demonstrate that for relatively large systems (> 100,000 atoms) the algorithm based on the hierarchy of wrapped spheres shows the best results and the traditional grid-based algorithm gives the worst timings. For small systems, however, the grid-based algorithm and the one based on the wrapped sphere hierarchy are beneficial.  相似文献

There is an increasing demand for quantitative data on metabolite exposure triggered by regulatory guidances. This contribution describes the accuracy of nanoelectrospray ionization mass spectrometry response of drug compounds and their metabolites from biological matrices compared with radiometric quantification. This is a comprehensive investigation of a set of real-life pharmaceutical compounds in relevant matrices such as urine, bile, feces and plasma. The data suggest that nanoelectrospray mass spectrometry can be used for semi-quantitation of metabolites in the absence of reference standards. Therefore, this approach is suitable to screen out non-relevant metabolites early in development as long as an adequate analytical error margin is applied thus balancing risks and resources.  相似文献

以农产品为原料的轻工业大都是典型的流程工业,在通过转化过程将原料转化为食品、饮料、添加剂、调味料、纸和中成药等产品的同时产生被称为过程残渣的固体废物与废料,如白酒糟、酒精糟、醋糟、甘蔗渣、中药渣、油粕、酱渣、菌渣和造纸黑液可熔渣等.这些残渣产生于特定的生产过程,富含纤维素、蛋白质或木质素,因此代表一种已经被集中的生物质资源.它们同时含水50%-80%、易腐烂变质、甚至呈弱酸碱性,因此是重要的环境污染源.本文着眼于轻工生物质过程残渣的高值化利用,分析指出富含纤维素的白酒糟、醋糟、甘蔗渣、中药渣、茶渣和造纸边角料等适合作为生物质能源而被转化利用,并根据资源特征提出了可能的技术路线.通过分别对热化学路线涉及的脱水干燥、燃烧发电与气化发电技术和集成乙醇发酵、沼气发酵的复合转化技术进行技术综述,最后针对不同规模的富含纤维素轻工生物质过程残渣能源化提供了技术选择建议.  相似文献

This paper describes the development and validation of a sequential injection (SI) anodic stripping voltammetry (ASV) method using the hanging mercury drop electrode for accumulation of the heavy metal cations Cu(II), Pb(II) and Cd(II). The method was applied to wastewater samples after proper acid digestion in open vessels to eliminate matrix effects. For a deposition time of 90 s at the flow rate of 10 μl s−1, the detection limits of the method were 0.06, 0.09 and 0.16 μmol L−1 for Cd, Pb and Cu, respectively. Under these conditions the linear dynamic range was between 0.20 and 9.0 μmol L−1 and the sampling frequency was 30 analyses per hour. The relative standard deviation of the method was 3%(n=7) at the concentration level of 2.0 μmol L−1. The accuracy of the method was evaluated by spiking the samples with known amounts of the metal cations, and by comparison with an independent analytical technique, the inductively coupled plasma atomic emission spectroscopy (ICP-AES). Average recoveries were around of 84%, and the results showed no evidence of systematic errors in comparison to the ICP-AES.  相似文献

This paper describes a sensitive spectrophotometric method for the determination of mercury, based on the flotation of a mercury iodide-brilliant green complex with cyclohexane at pH 5 and dissolution of the complex in acetone. The absorbance of the complex is measured at 625 nm. Beer's law is obeyed from 4–500 g/l of mercury. The method is highly sensitive ( = 5.96 × 105 1 mol–1 cm–1) and selective, as it is free from interferences by almost all cations and anions in the presence of EDTA. Furthermore, a two-level orthogonal array design as a chemometric method has been used for optimization of the various parameters involved. The method has been successfully applied to paper industry effluents.  相似文献

Thermal analysis methods are well-established techniques in research laboratories of pharmaceutical industry. The robustness and sensitivity of instrumentation, the introduction of automation and of reliable software according to the industrial needs widened considerably the areas of applications in the last decade. Calibration of instruments and validation of results follow the state of the art of cGMP as for other analytical techniques. Thermal analysis techniques are especially useful for the study of the behavior of the poly-phasic systems drug substances and excipients and find a unique place for new delivery systems. Since change of temperature and moisture occur by processing and storage, changes of the solid state may have a considerable effect on activity, toxicity and stability of compounds. Current requirements of the International Conference of Harmonisation for the characterization and the quantitation of polymorphism in new entities re-enforce the position of thermal analysis techniques. This challenging task needs the use of complementary methods. Combined techniques and microcalorimetry demonstrate their advantages. This article reviews the current use of thermal analysis and combined techniques in research and development and in production. The advantage of commercially coupled techniques to thermogravimetry is emphasized with some examples. The aim of this work is development of methodology for analysis of inorganic cations (sodium, ammonium, potassium, magnesium and calcium) in fertilizer industry wastewater. Method development includes optimization of eluent flow rate and concentration of eluent competing ion in order to obtain optimal separation within reasonable analysis time. For that purpose artificial neural network retention model was developed and used in combination with normalized resolution product criteria function. Developed artificial neural network retention model shows good predictive ability R2 ≥ 0.9983. The determined ion chromatographic parameters enable baseline separation of all components of interest. By performing validation procedure and number of statistical tests it is shown that developed ion chromatographic method has superior performance characteristic: linearity R2 ≥ 0.9984, recovery = 99.81% − 99.44%, repeatability RSD ≤ 0.52%. That result proves that proposed method can be used for routine monitoring analysis in fertilizer industry.  相似文献

A hypergraph is simple if it has no two edges sharing more than a single vertex. It is s‐list colorable (or s‐choosable) if for any assignment of a list of s colors to each of its vertices, there is a vertex coloring assigning to each vertex a color from its list, so that no edge is monochromatic. We prove that for every positive integer r, there is a function dr(s) such that no r‐uniform simple hypergraph with average degree at least dr(s) is s‐list‐colorable. This extends a similar result for graphs, due to the first author, but does not give as good estimates of dr(s) as are known for d2(s), since our proof only shows that for each fixed r ≥ 2, dr(s) ≤ 2 We use the result to prove that for any finite set of points X in the plane, and for any finite integer s, one can assign a list of s distinct colors to each point of the plane so that any coloring of the plane that colors each point by a color from its list contains a monochromatic isometric copy of X. © 2011 Wiley Periodicals, Inc.
